PEG 200 is a clear liquid at room temperature used as a solvent, plasticizer, and moisture absorber in liquid formulations. PEG 4000 and 6000 are solid waxy materials at room temperature, used as binders, bases for suppositories, and release modifiers in solid dosage forms. As molecular weight increases, melting point, viscosity, and crystalline content increase while solubility and hygroscopicity decrease. The physical form determines the application - liquid for solutions, solid for tablets and topical bases.

Low molecular weight PEGs (200-600) serve as plasticizers, solvents, and humectants in oral and topical liquids. Medium molecular weight PEGs (1000-2000) are used as ointment bases and suppository vehicles. High molecular weight PEGs (3350-8000) function as tablet binders, release retardants, and mucoadhesive polymers. PEG 4000 is commonly used in solid dispersions to improve drug solubility, while PEG 6000 is preferred as a sustained-release matrix former.

PEG stabilizes protein and peptide therapeutics through preferential exclusion and steric stabilization. PEG molecules are preferentially excluded from the protein surface, creating a thermodynamic driving force for the protein to remain in its folded, native state. The PEG polymer chain also forms a protective steric barrier around the protein, preventing aggregation, surface adsorption, and proteolytic degradation. This PEGylation effect has made PEG the gold standard for stabilizing biologic drugs.

PEG is generally recognized as safe for injectable formulations at approved levels. PEG 300 and 400 are used as co-solvents in parenteral products at concentrations typically below 50% v/v. PEG 4000 and 6000 are used at lower concentrations (<10%) in injectable sustained-release systems. The WHO and FDA have established acceptable daily intake levels, and PEG has a well-documented safety profile. However, high-molecular-weight PEGs should be avoided in renally impaired patients.
